Why not experiment? If you prefer not to use flash, the other suggestion I'd have is to crank your 350D up to ISO 1600 or so and try shooting at f/4 or f/5.6 in fairly bright light with your 100mm macro. You could use a desk lamp or bright, concentrated lamp to get more light onto the eye. Shooting ...
