Hello graham and the others... :-)
With its 300 spiecies, the genus Spirogyra is very difficult to identifie... thats depend on the number of chloroplast and the type of "Partition" .
some times as in this picture, there is only a few chloroplast that give the oportunity to well observe the nucleus wich is as you said in the middle of the cell.

I try to obtain an home made DIC with some wollaston on my old Ortholux 1... the photo is not as good as I hope but the nucleus is clearly visible with the cystoplasmic strands...

I am realy sorry but I remember seeing some marvellous images of spyrogyra in DIC from some one on this forum but I am not able to remember who he is....

Hi Dominique,

Well, I have to say your home made DIC is looking really good. I have done a little experimenting along these lines too, but not with nearly the results you are obtaining.

Really great photo in my opinion. Shows great detail of the nucleus and surrounding structure. Makes me want to give it another go.

Where in Normandy do you live?

- nice image Dominique. Apparantly of the 400 or so species of Spirogyra, about 50 have been reported in the UK

- your home made DIC sounds fascinating and looks like it is giving some good results.
